Enzyme cellulase có nguồn gốc từ vi khuẩn cellulolytic ngày càng được quan tâm nhờ tiềm năng ứng dụng trong công nghệ sinh học. Nghiên cứu này nhằm đánh giá các yếu tố điều kiện nuôi cấy ảnh hưởng đến hoạt tính cellulase của chủng Bacillus velezensis DSC.04 bản địa. Chủng vi khuẩn được nuôi trong môi trường CB với các điều kiện khác nhau gồm pH ban đầu, nhiệt độ ủ, nồng độ NaCl, nồng độ CMC, thời gian ủ và thể tích chất cấy. Dịch enzyme thô thu nhận được sử dụng để xác định hoạt tính Carboxymethyl Cellulase (CMCase, U/mL) bằng thuốc thử DNS. Kết quả cho thấy tất cả các yếu tố nuôi cấy đều ảnh hưởng đến hoạt tính CMCase của chủng DSC.04. Phân tích mô hình Plackett-Burman xác định 4 yếu tố có tác động chính đến hoạt tính CMCase gồm pH, nồng độ NaCl, nồng độ CMC và tỷ lệ chất cấy. Điều kiện nuôi cấy thích hợp cho quá trình sinh tổng hợp cellulase của chủng DSC.04 được xác định là pH 7, bổ sung 1,5% NaCl (w/v), 1% CMC (w/v), 2% dịch cấy (v/v), ủ ở 35°C trong 48 giờ với hoạt tính CMCase đạt 1,62 U/mL.
