DETERMINATION OF MOLECULAR MARKERS AND REGENERATION OF IN VITRO SHOOTS OF Polygonatum kingianum Coll ex Hemsl.

Polygonatum kingianum Coll. Et Hemsl. is an important medicinal material in traditional medicine . In this study, Explants of were collected from SaPa-Lao Cai and classified at species level through anatomical characteristics and two molecular markers of this species including psbA-trnH and rpoC1 were sequencing. After, in vitro shoots of P. kingianum were regenerated from young shoots of rhizome for multiplication to conserve this species. The result shows that the first sequence, psbA-trnH, has 616 bp and is a good marker for identification at species level. The second sequence, rpoC1, has 542 bp and is useful to classify Polygonatum genus. Young shoots of rhizome were suface sterilized in 70% (v/v) ethanol for 15 minutes, in 10% (v/v) javel (Sodium hypochlorite) solution for 15 minutes. The medium including MS, 30 g/l saccharose, 7 g/l agar and supplemented with 5 mg/l BAP and 0.2 mg/l IAA was suitable for in vitro shoots regeneration. For the first time, in vitro shoots regeneration of Polygonatum kingianum Coll. Et Hemsl. was established , this result is potential for convervation medicinal material in the future.
