Abstract

The aim of this study was to establish a protocol for in vitro propagation of Schisandra sphenanthera from juvenile stem segments containing dormant buds. The results indicated MS minerals were more effective than WPM ones in in vitro regeneration culture, and MS+ 1 mg/L BAP + 30 g/L sucrose was the best with a regenerated percentage of 93.33% after 12 weeks of culture. The highest shoot multiplication coefficient was recorded on the MS supplemented with 1 mg/L BA + 0.5 mg/L IBA + 30 g/L sucrose, reaching 8.93 shoots with 3,75cm in height. The quarter-strength MS medium augmented with 0.75 mg/L IAA and 20 g/L sucrose was the best medium for in vitro rooting of S. sphenanthera shoots, giving a rooting percentage of 86,67% with 4.82 roots/shoot. This research result is the initial in propagating of S. spenanthera, and also the first report for species belonging to the genus Schisandra in Viet Nam, contributing to preserving the rare genetic resources and unlocking the potential for sustainable development of products from this medicinal plant.
