ANALYSIS OF THE GENOTYPE VALUE OF MULTI-TOED CHICKEN OF THE DAO PEOPLE IN THAI NGUYEN PROVINCE

Abstract


The multi‐toed chicken (also known as multi-spurred chickens) of the Dao ethnic minority in Thai Nguyen represents a valuable genetic resource with strong potential for selective breeding due to its meat quality. The FGFBP2 gene, which is involved in muscle and adipose tissue development, was investigated to assess the genetic characteristics of this breed. In 2021, thirty blood samples from multi‐toed chickens were collected in Thai Nguyen province. Total DNA was extracted and the FGFBP2 gene was amplified by PCR. PCR products were purified, and ten representative samples were sequenced using an ABI PRISM 3100. Results showed that all thirty samples possessed the FGFBP2 gene, with nucleotide similarity among the ten sequenced samples ranging from 99.3 to 100% and amino acid similarity of 100%. When compared with twenty reference sequences, nucleotide similarity ranged from 83.1 to 99.7%, with the highest similarity to Southeast Asian red junglefowl. A neighbor-joining phylogenetic tree indicated that the multi-toed chicken breed clusters closely with species in the Phasianidae family. This study confirms the genetic distinctiveness and conservation value of the multi-toed chicken breed and provides a scientific basis for its future conservation and breeding programs.
