STUDY ON THE ADVENTITIOUS ROOT PROLIFERATION OF Codonopsis javanica IN LIQUID SHAKING CULTURE

Abstract


Codonopsis javanica is a medicinal plant that contains various valuable bioactive compounds. This study was conducted to obtain the adventitious root biomass of C. javanica in liquid shaking culture. Node explants derived from in vitro shoots were cultured on solid medium to induce adventitious roots. They were subsequently proliferated in a liquid shaking culture system. Several factors affecting the proliferation of adventitious roots in liquid shaking culture were investigated, including plant growth regulators, mineral medium, light conditions, and culture time. The results showed that the optimal medium for the proliferation of adventitious roots in the liquid shaking culture system was SH medium supplemented with 0.5 mg/L IBA, 30 g/L sucrose, pH 5.8, and the culture was kept in dark conditions. The highest proliferation rate and polysaccharide accumulation of adventitious roots were achieved on the 35th day of culture, with a multiplication coefficient of 4.3 times and polysaccharide content of 13.98%. Qualitative analysis of the chemical composition revealed that the adventitious root biomass of C. javanica contained bioactive compounds including alkaloids, carbohydrates, phenolics, flavonoids, oils, and fats. These findings refer to the large-scale commercial production of C. javanica root biomass using bioreactor systems, which offer a promising alternative to traditional cultivation methods.
