REGENERATION OF PAPHIOPEDILUM CONCOLOR FROM YOUNG BUDS AND SEEDS

Abstract


Paphiopedillum concolor is one of the endemic species of Vietnam. It is a small slipper orchird with relatively large pale yellowish fragrant flowers finely spotted with purple. It has both aesthetic and medicinal value. Some of scientists  has been reporded the result of  research  on the diversity, regeneration or propagation of Paphiopedilum concolor from auxilary buds. This research was conducted to investigate in vitro propagation of Paphiopedillum concolor collected in Thainguyen, Vietnam. Materials for in vitro propagation include of young buds and seeds collected from fruits aged about 170-200 days after pollination (DAP). In conclusion, young buds sterilized by 0.1% HgCl2 for 15 minutes for a 50% clean sample rate after two weeks. In vitro shoots grew and developed well, leaf number per shoot reached 3.8 and leaf width reached 0.70 cm after four months. P. concolor fruits aged about 170 days after pollination (DAP) sterilized by  0.1% HgCl2 solution for 15 minutes for a clean sample rate of 87,5%. Seeds germinated on MS medium supplemented with BAP 2mg/l (Seed germination rate reached 81,7%). Seed germination time was 90 days to 100 days. Shoots formed 1 to 2 leaves after 90 days of culture. In vitro P. concolor plantlets have been created with 2- 3 leaves, 1-2 small and short roots after 110-120 days cultivaion. This reseach has created materials to complete the protocol of propagation of Paphiopedillum concolor.
