PRODUCTION AND BIOACTIVITY ASSESSMENT OF EXOPOLYSACCHARIDE PRODUCED BY LEVILACTOBACILLUS BREVIS MT8

Abstract


Expolysaccharides produced by lactic acid bacteria have gained great interest for their potential applications in food, pharmaceutical and biotechnological industries due to their biological and rheological properties. In Vietnam, a few studies have explored production and bioactivity of lactic acid bacteria-derived EPS. The present study reports the improvement of EPS production by Levilactobacillus brevis MT8, in addition to assessing its antioxidant and antibacterial activities. The isolate MT8 from fermented eggplant, showing mucoid phenotype, was identified as L. brevis using 16S rRNA gene analysis. After optimization, the strain MT8 produced the maximum EPS yield of 725.7±50.1 mg/L when cultivated in the MRS medium for 48 h at 37°C, in which 20 g/L glucose was replaced by 40 g/L sucrose. Antioxidant assay revealed that the obtained EPS exhibited the highest scavenging activities against 1,1-diphemy 1-2-picryl-hydrazyl (78.0±0.4%) and hydroxyl (68.8±0.5%) at 2 mg/mL. Of note, 5 mg/mL EPS also showed antibacterial activity against Escherichia coli ATCC 11105 and Salmonella Typhimurium ATCC 14028 with inhibition zones of 5.9±0.4 mm and 7.5±0.2 mm, respectively. Therefore, the EPS synthesized by L. brevis MT8 shows promise for application in the food, cosmetics, biomedical industries. To the best of our knowledge, this is the first report to prove antibacterial activity of EPS produced by L. brevis.
