Abstract


Phytophthora palmivora is the only currently recorded microbial disease causing death of durian trees. The study conducted the isolation, selection and cultivation of Endophytic bacterial strains with potential antifungal effect against P. palmivora. Using the method of isolating Endophytic bacteria from 12 durian samples in Cu Kuin district, 29 strains were cloned. The antagonistic activity of these Endophytic bacteria against P. palmivora was carried out using the co-culture method in vitro. Eleven strains were selected that have the ability to resist P. palmivora with an antagonistic effect around 50.62 - 67.41%, the EB.CK9 strain with the highest resistance to P. palmivora (67.41%) was identified as Bacillus amyloliquefaciens based on the 16S rRNA gene sequencing method. Results of propagation in a conical flask showed that strain EB.CK9 grew best at 36oC, pH 7, 200rpm, after 48 hours it reached 44.5 x108 CFU/ml. The cultivation was further conducted in a 14 L Bioreactor system, and the EB.CK9 strain reached a higher density of 8.25x1011 CFU/ml in a shorter cutivation time (10 hours) comparing to cultivation in flaks. The research opens up application directions for developing indigenous biological products from Endophytic bacteria for effective and sustainable cultivation of durian.
