Abstract


Mokara is a hybrid orchid derived from three genetically different genera Arachnis, Ascocentrum and Vanda. This hybrid orchid is one of the important flower cuttings and is widely grown in many countries such as Singapore, Indonesia, Thailand... and Vietnam. This research aims to micropropagate Mokara orchid from leaf tissue through PLB generation. The results of this study showed that MS medium supplemented with 2.0 mg l-1 2.4-D was suitable for inducing PLB from leaf tissue. The best shoot formation was obsserved in MS medium supplemented with 10% coconut water and 2.0 mg l-1 BAP. In vitro rooting was best suited on MS medium supplemented with 1.0 mg l-1 NAA. These results could be the basis for building an in vitro propagation process for Mokara orchid which could applied in the production of this valuable orchid variety.
