Abstract


In this study, a procedure for quantifying α-mangostin in mangosteen peel by high performance liquid chromatography (HPLC) with a PDA detector was carried out. Chromatographic separation was achieved using a symmetry C18 column (4.6 mm x 250 mm) with the mobile phase consisting of acetonitril : water (95 : 5, v/v) at a flow rate of 1.5 ml/min, and monitored at 320 nm. The method was validated in accordance with International Conference on Harmonisation (ICH) guidelines. The calibration curve was linear over the concentration range from 0.01 to 200 µg/ml. The limit of detection (LOD) was 0.01 µg/ml and the limit of quantitation (LOQ) was 0.03 µg/ml. The method achieved accuracy with recovery in the range of 99.2 – 101.9%, high repeatability with an RSD of 0.746 % for peak area and 0.074 % for retention time. The procedure was applied to determine the α-mangostin content in mangosteen peels of several different regions in Viet Nam.
