DETERMINATION OF BERBERIN CONTENT IN TINOSPORA SINENSIS COLLECTED IN SAN HO DISTRICT, LAI CHAU PROVINCE BY HIGH-PERFORMANCE CHROMATOGRAPHIC METHOD

This study aims to determine the berberine content in the medicinal herb Tinospora sinensis (Lour) Merr. Menispermaceae were collected in Sin Ho district, Lai Chau province by high performance liquid chromatography. In the study, the medicinal plant sample was extracted using the soxhlet technique with methanol solvent, and concentrated extract to make a high liquid by vacuum rotary evaporator. The content of berberine in the medicinal liquid was quantified by the HPLC method with the condition of reversed-phase column (C18, 250 mm x 4.6 mm, 5µm), the water-acetonitrile mobile phase at a ratio of 1-1 (for every 1000 ml of solvent added 3.4 g potassium dihydro phosphate and 1.7 g sodium lauryl sulfate), sample injection volume 10 µl, flow rate 1.0 ml/min, detection wavelength 345 nm. The analytical method was evaluated for the following criteria: systematic suitability, specificity, linearity, accuracy, accuracy, LOD, and LOQ. The study obtained results of a berberine peak with a retention time of about 9.8 minutes. Quantitative methods meet the requirements of the system's specificity and suitability; The accuracy has an average refind rate of 99.98% with a relative standard deviation of 8.46%; Linear ranges were constructed with berberine concentrations from 5 µg/ml to 80 µg/ml with a correlation coefficient R2 = 0.9957. Applying the quantitative method, the berberine content in the research sample was determined to be 0.162 ± 0.011 mg/g.
