Abstract


Paphiopedilum emersonii Koop & P.J. Cribb has a very narrow distribution in Vietnam, only found in some provinces such as Tuyen Quang, Thai Nguyen, Bac Kan, and Cao Bang. Paphiopedilum emersonii (P. emersonii) is a very popular flower for international visitors, so this species is increasingly exploited and threatened with extinction. Therefore, the study of in vitro propagation of P. emersonii is very essential. In this study, the fruit of P. emersonii was used as a sterilization material and non-symbiotic germination on different basic culture media to produce in vitro shoots. We have successfully studied a number of factors affectting the germination ability of P. emersonii. The results showed that fruits can be disinfected with 2% NaOCl solution for 30 minutes, the clean sample rate reaches 85% after 30 days. The ability of seed germination depends on the age of fruit and basic media. The suitable fruit age for germination is about 180 days after pollination. The suitable medium is ½ MS or ½ VW environment, germination time is 90 - 100 days. The germinated seeds developed protocorm and differentiated well into shoots on MS medium supplemented with 1.0 mg/L BAP, 0.5 mg/L NAA and 100 ml/L coconut water, 40 g/L potatoes homogeneous, banana homogeneous 30 g/L. These results are a premise for in vitro breeding studies of P. emersonii in Vietnam.
